Overview

NGM438 + pembrolizumab is a combination investigational regimen under clinical evaluation for the treatment of advanced or metastatic solid tumors. NGM438 is a monoclonal antibody antagonist of Leukocyte-associated immunoglobulin-like receptor 1 (LAIR1), designed to reverse collagen-driven immune suppression in the tumor microenvironment. Pembrolizumab is an approved humanized monoclonal antibody immune checkpoint inhibitor of programmed cell death protein 1 (PD-1), marketed as Keytruda. The rationale for the combination is that LAIR1 antagonism with NGM438 may remodel immunosuppressive stromal environments and sensitize tumors resistant to PD-1 blockade, augmenting the anti-tumor efficacy of pembrolizumab.
