NGM707 + pembrolizumab is an investigational combination therapy for advanced or metastatic solid tumors. NGM707 is a novel, fully human monoclonal antibody that acts as a dual antagonist of Immunoglobulin-like transcript 2 (ILT2) and Immunoglobulin-like transcript 4 (ILT4), two inhibitory receptors expressed on myeloid cells and some lymphoid cells. By blocking both ILT2 and ILT4, NGM707 aims to reprogram the tumor microenvironment to enhance anti-tumor immune responses. Pembrolizumab (Keytruda) is an approved anti-PD-1 monoclonal antibody that blocks the Programmed cell death protein 1 (PD-1) receptor on T cells, thereby promoting immune-mediated tumor cell killing. The combination seeks to overcome resistance mechanisms in tumors unresponsive to PD-(L)1 blockade alone by targeting both myeloid and lymphoid immunosuppressive pathways. Early clinical data show the combination is generally well tolerated with preliminary evidence of efficacy in heavily pretreated patients with advanced solid tumors, including microsatellite stable colorectal cancer (MSS CRC), which typically does not respond to PD-(L)1 monotherapy[1][3][5][6].
