Overview

**Ni(II) proline dithiocarbamate** is a small molecule metal complex composed of a nickel(II) ion chelated by a dithiocarbamate ligand derived from the amino acid proline. It is designed as a potential anticancer agent, particularly for breast cancer. Mechanistically, the complex is thought to exert its cytotoxic effect by interacting with biological targets such as the estrogen receptor alpha (ESR1), disrupting essential protein functions involved in cell survival and apoptosis. Molecular docking studies indicate binding to the estradiol-estrogen receptor-α, while in vitro studies on the MCF-7 breast cancer cell line demonstrate cytotoxicity with an IC50 of approximately 315.70 µg/mL[2][3]. The compound is under investigation as a promising chemotherapeutic agent with minimal systemic toxicity, aided by its design for improved selectivity, cellular uptake, and water solubility[2][3].
