Overview

NI-1801 is a bispecific monoclonal antibody that targets both CD47 and mesothelin. By binding to CD47, it blocks the interaction with signal regulatory protein alpha (SIRPalpha), an inhibitory protein on macrophages, thereby promoting phagocytosis of tumor cells. The mesothelin targeting component directs the antibody to tumors expressing this antigen. Paclitaxel is a small molecule chemotherapeutic agent classified as a taxane; it stabilizes microtubules and prevents their depolymerization, disrupting mitosis and leading to cell death. The combination of NI-1801 with paclitaxel is being investigated for enhanced antitumor activity in patients with advanced, metastatic, or recurrent solid malignancies expressing mesothelin[3][5][6].
