Niclosamide + camostat is a combination of two small molecule drugs. Niclosamide is an anthelmintic agent primarily used to treat tapeworm (cestode) infections in humans. It acts by disrupting oxidative phosphorylation in the mitochondria of parasites, leading to ATP depletion and death of the worm. Beyond its antiparasitic use, niclosamide has demonstrated activity against various signaling pathways (such as Wnt/β-catenin, mTORC1, STAT3, NF-κB, and Notch), suggesting potential utility in cancer and viral infections[1][2][6]. Camostat is a serine protease inhibitor most commonly used for chronic pancreatitis and has been investigated for its ability to inhibit TMPRSS2—a host cell protease involved in viral entry—making it a candidate for antiviral therapy including COVID-19. The combination has been explored experimentally as a potential antiviral regimen due to their complementary mechanisms targeting both viral entry (camostat) and replication or host response pathways (niclosamide).
