This is a combination of two drugs: nicotine, a well-known nicotinic acetylcholine receptor agonist and stimulant, and AVL-3288, an investigational small molecule that acts as a selective type I positive allosteric modulator (PAM) of the alpha7 nicotinic acetylcholine receptor (α7 nAChR). Nicotine directly activates various subtypes of nicotinic receptors in the central and peripheral nervous systems. AVL-3288 does not activate α7 nAChRs directly but enhances their response to endogenous agonists like acetylcholine or exogenous agents such as nicotine. The rationale for combining these agents is to potentiate cholinergic neurotransmission via α7 nAChR modulation, which may have therapeutic potential for cognitive deficits associated with disorders such as schizophrenia or traumatic brain injury. Preclinical studies show that positive allosteric modulation of α7 nAChRs can improve cognitive function; clinical trials with AVL-3288 alone have shown it to be safe and well-tolerated but with limited efficacy signals in early-phase studies[2][3][5]. Trials involving both drugs together are designed to assess whether concurrent administration alters safety or neurocognitive effects compared to either agent alone[2][4].
