Nicotine + galantamine is an experimental combination of two small molecule drugs, nicotine and galantamine. Nicotine is a nicotinic acetylcholine receptor (nAChR) agonist, while galantamine is both a reversible acetylcholinesterase inhibitor and a positive allosteric modulator of nAChRs. The combination has been shown in preclinical studies to have synergistic neuroprotective effects, particularly through enhanced activation of the alpha7 nicotinic acetylcholine receptor (alpha7 nAChR). This synergy results in greater inhibition of microglial activation and increased protection against dopaminergic neuronal loss compared to either agent alone[1][2]. Galantamine's dual action increases synaptic acetylcholine levels and potentiates the effect of nicotine at nAChRs, especially alpha7 subtypes. The combination has been investigated for potential therapeutic use in neurodegenerative diseases such as Parkinson's disease and HIV-associated dementia due to its anti-inflammatory and neuroprotective properties[1][2].
