This pharmacological combination consists of nicotine polacrilex, a nicotine replacement therapy (NRT), and bupropion, an atypical antidepressant. Nicotine polacrilex is a resin complex that delivers a controlled dose of nicotine to the systemic circulation via the buccal mucosa, acting as an agonist at nicotinic acetylcholine receptors to alleviate withdrawal symptoms. Bupropion functions as a norepinephrine-dopamine reuptake inhibitor (NDRI) and also acts as a non-competitive antagonist at neuronal nicotinic acetylcholine receptors. This dual action helps to reduce the urge to smoke and diminishes the reinforcing effects of nicotine. The combination was specifically evaluated by the University of California, San Francisco (UCSF) in clinical trials (such as NCT00086385) aimed at maintaining smoking cessation and preventing relapse in older adults (aged 50 and older) through extended pharmacological support and tailored counseling.
