Overview

Nilotinib + cetuximab is an investigational combination therapy involving two distinct anticancer agents. Nilotinib is a small molecule tyrosine kinase inhibitor primarily targeting BCR-ABL, KIT, and PDGFR kinases. It is approved for the treatment of Philadelphia chromosome-positive chronic myeloid leukemia (CML), including cases resistant to imatinib. Cetuximab is a monoclonal antibody that targets the epidermal growth factor receptor (EGFR) and is approved for use in metastatic colorectal cancer and head and neck squamous cell carcinoma. The rationale for combining these drugs lies in their complementary mechanisms—nilotinib inhibits intracellular signaling pathways critical to cancer cell proliferation, while cetuximab blocks EGFR-mediated signaling at the cell surface. This combination has been studied in early-phase clinical trials to determine safety and efficacy in solid tumors[2][3][6].
