Overview

Nilotinib + paclitaxel is an investigational combination therapy consisting of two small molecule drugs used together for the treatment of advanced solid tumors, particularly in patients who have progressed after prior taxane-based chemotherapy. Nilotinib is a BCR-ABL tyrosine kinase inhibitor that blocks signaling pathways involved in cancer cell proliferation and survival. Paclitaxel is a microtubule-stabilizing agent that disrupts mitosis, leading to cell death. The rationale for combining these agents includes potential synergistic anticancer effects and evidence suggesting that nilotinib may reduce the incidence or severity of peripheral neuropathy caused by paclitaxel. Early-phase clinical trials have demonstrated partial responses and durable disease control in select patients with relapsed or refractory solid tumors, including ovarian granulosa cell tumors and endometrial carcinoma[1][2][5][7]. The combination is being further evaluated as part of precision medicine initiatives such as ComboMATCH.
