Nimacimab + semaglutide is an investigational combination therapy for obesity and overweight with weight-related comorbidities. Nimacimab is a first-in-class, peripherally-restricted monoclonal antibody that acts as a negative allosteric modulator (inverse agonist/antagonist) of the cannabinoid receptor type 1 (CB1). Semaglutide is a glucagon-like peptide-1 (GLP-1) receptor agonist, already approved for type 2 diabetes and chronic weight management. The combination aims to leverage complementary mechanisms—CB1 inhibition by nimacimab to promote fat loss while preserving lean mass, and GLP-1 receptor activation by semaglutide to reduce appetite and improve glycemic control. Early clinical studies are evaluating the safety, tolerability, and efficacy of this combination in producing greater weight loss than either agent alone[2][4][5][7].
