The treatment regimen known as CCRT-Sun Yat-sen University-nasopharyngeal carcinoma involves the combination of **nimotuzumab**, a humanized monoclonal antibody, with concurrent chemoradiotherapy (CCRT) consisting of cisplatin and intensity-modulated radiotherapy (IMRT). Nimotuzumab specifically targets the epidermal growth factor receptor (EGFR), a protein often overexpressed in nasopharyngeal carcinoma (NPC) that promotes tumor cell proliferation, survival, and resistance to therapy. By binding to the extracellular domain of EGFR, nimotuzumab blocks ligand-induced activation and downstream signaling pathways such as PI3K/Akt and MAPK. This inhibition enhances the cytotoxic effects of cisplatin-induced DNA damage and ionizing radiation. This specific combination is being investigated by the Sun Yat-sen University Cancer Center for patients with high-risk, locoregionally advanced NPC, particularly those who exhibit a suboptimal response to induction chemotherapy as evidenced by persistent Epstein-Barr virus (EBV) DNA levels.
