Nimotuzumab + temozolomide is a combination therapy used primarily in the treatment of glioblastoma multiforme (GBM) and other high-grade gliomas. Nimotuzumab is a humanized monoclonal antibody that targets the epidermal growth factor receptor (EGFR), inhibiting its signaling and thereby reducing tumor cell proliferation, survival, and angiogenesis. Temozolomide is an oral alkylating agent that induces DNA damage in tumor cells, leading to apoptosis. The combination has shown enhanced antitumor activity compared to either agent alone, particularly in tumors with EGFR overexpression or EGFRvIII mutation. Clinical studies indicate this regimen can improve progression-free survival and overall survival when added to standard radiotherapy protocols for newly diagnosed GBM patients[1][3][5][6]. The safety profile of the combination is generally manageable.
