Overview

Nimustine + teniposide is a combination chemotherapy regimen used primarily in the treatment of malignant gliomas, including glioblastoma. Nimustine (ACNU) is a nitrosourea alkylating agent that works by cross-linking DNA strands and inhibiting DNA replication and transcription. Teniposide (VM26) is a topoisomerase II inhibitor that induces DNA strand breaks by stabilizing the enzyme-DNA complex during replication. The combination has been studied as both first-line and salvage therapy for glioblastoma, particularly after failure of standard temozolomide-based regimens. Clinical studies indicate moderate efficacy but significant hematologic toxicity, especially myelosuppression[1][2][3][6].
