Overview

Combination of **nintedanib** and **pirfenidone**, two orally administered small molecule antifibrotic agents primarily used in the treatment of idiopathic pulmonary fibrosis (IPF). Nintedanib acts as a **tyrosine kinase inhibitor** targeting receptors involved in fibrogenesis, including PDGF, FGF, and VEGF receptors, while pirfenidone modulates TGF-beta signaling and inhibits fibroblast proliferation. This combination is being explored for additive or synergistic effects in decelerating IPF progression. Clinical studies indicate the combo is generally safe, though gastrointestinal side effects are common, and tolerability is often limited by adverse events. There is emerging evidence for potential use in other fibrosing interstitial lung diseases, including silicosis[1][2][3][4].
