Overview

Nipocalimab + hydroxychloroquine is a **combination therapy** being explored for the treatment of autoimmune and inflammatory diseases. **Nipocalimab** is a monoclonal antibody designed to bind with high affinity to the neonatal Fc receptor (FcRn), thereby blocking FcRn-mediated recycling of IgG and reducing circulating pathogenic IgG autoantibodies. **Hydroxychloroquine** is an immunomodulatory small molecule that raises lysosomal pH and impacts antigen presentation and inflammatory pathways. This combination aims to target complementary immune functions, with nipocalimab lowering pathogenic IgG and hydroxychloroquine modulating the underlying autoimmune response. Preclinical and phase 1 data explored pharmacokinetic and pharmacodynamic interactions; hydroxychloroquine does not significantly affect the IgG-lowering effect or pharmacokinetics of nipocalimab, and both are generally well tolerated when co-administered[1][2].
