Overview

Niraparib + bevacizumab is a combination therapy used primarily as maintenance treatment in advanced ovarian cancer. Niraparib is an orally active small molecule inhibitor of poly(ADP-ribose) polymerase (PARP), specifically targeting PARP-1 and PARP-2, which are enzymes involved in DNA repair. By inhibiting these enzymes, niraparib induces synthetic lethality in cancer cells with homologous recombination deficiency, such as those with BRCA mutations[3][6]. Bevacizumab is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis and reducing tumor blood supply[4]. The combination has shown improved progression-free survival and durable overall survival without cumulative toxicity in patients with newly diagnosed advanced ovarian cancer who have responded to first-line platinum-based chemotherapy plus bevacizumab[1][2][8].
