Overview

Niraparib + sintilimab is an investigational combination therapy consisting of two agents: niraparib, a small molecule poly(ADP-ribose) polymerase (PARP) inhibitor, and sintilimab, a monoclonal antibody targeting programmed cell death protein 1 (PD-1). Niraparib inhibits PARP enzymes involved in DNA repair, leading to synthetic lethality in tumor cells with homologous recombination repair deficiencies. Sintilimab blocks the PD-1 immune checkpoint pathway, enhancing T-cell mediated anti-tumor immunity. The combination aims to exploit potential synergistic effects between DNA damage induction and immune checkpoint blockade. This regimen is under clinical investigation for various advanced solid tumors including recurrent/metastatic nasopharyngeal carcinoma, endometrial cancer, non-small cell lung cancer with HRR gene mutations, and other malignancies[1][2][3][5].
