Overview

Niraparib + toripalimab is an investigational combination therapy comprising two distinct agents with complementary mechanisms of action, evaluated primarily in oncology clinical trials. Niraparib is a small molecule inhibitor of poly(ADP-ribose) polymerase (PARP), which impairs DNA repair in cancer cells, leading to cell death—especially in tumors with homologous recombination deficiency. Toripalimab is a monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to enhance anti-tumor immune responses. The combination aims to exploit both direct tumoricidal effects and immunomodulation for synergistic efficacy. This regimen has been studied in phase II clinical trials for metastatic, recurrent, or persistent cervical cancer[1][2][3][4].
