NIS793 + gemcitabine + nab-paclitaxel is a combination regimen under investigation for the first-line treatment of metastatic pancreatic ductal adenocarcinoma. NIS793 is a fully human IgG2 monoclonal antibody that targets and inhibits the transforming growth factor beta (TGF-β) pathway, which is known to regulate the tumor microenvironment and promote immune exclusion. By inhibiting TGF-β, NIS793 is expected to augment the efficacy of chemotherapy and enhance immune response. Gemcitabine is a nucleoside analog chemotherapeutic, and nab-paclitaxel is a nanoparticle albumin-bound formulation of paclitaxel, both broadly used as standard-of-care drugs in pancreatic cancer. The combination is being studied to improve response rates and survival by targeting both tumor proliferation and stromal immune modulation[3][8].
