NIS793 + mFOLFIRINOX is an investigational combination therapy being evaluated for the treatment of pancreatic adenocarcinoma. NIS793 (letolizumab) is a fully human monoclonal antibody that targets and neutralizes transforming growth factor-beta (TGF-β), a cytokine often overexpressed in the tumor microenvironment that promotes immune evasion, fibrosis, and metastasis. mFOLFIRINOX is a standard-of-care chemotherapy regimen comprising oxaliplatin, irinotecan, leucovorin, and 5-fluorouracil. By combining TGF-β inhibition with cytotoxic chemotherapy, the regimen aims to overcome the dense stroma and immunosuppressive environment characteristic of pancreatic tumors, potentially improving surgical resectability and long-term outcomes in the neoadjuvant setting.
