Overview

NIS793 is a human IgG2 monoclonal antibody that binds to transforming growth factor beta (TGF-β), inhibiting its signaling pathway. Spartalizumab is a humanized IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1), acting as an immune checkpoint inhibitor. The combination of NIS793 and spartalizumab is being investigated for the treatment of advanced solid tumors, with the rationale that dual blockade of TGF-β and PD-1 may enhance antitumor immunity by overcoming immunosuppressive mechanisms in the tumor microenvironment[2][5]. Clinical trials have shown that this combination is well tolerated, with no dose-limiting toxicities observed during dose escalation. Biomarker analyses support proof of mechanism for TGF-β inhibition and modulation of immune signatures in tumor biopsies[2][5].
