Overview

This drug regimen is a combination therapy consisting of: - NIS793, a fully human monoclonal antibody targeting transforming growth factor beta (TGF-β), developed as an immunomodulatory antineoplastic agent to inhibit TGF-β signaling within the tumor microenvironment[7][8]. - Tislelizumab, a humanized IgG4 monoclonal antibody against programmed cell death protein 1 (PD-1), primarily used as an immune checkpoint inhibitor in various solid tumors. - Bevacizumab (brand: Avastin), a humanized monoclonal antibody inhibiting vascular endothelial growth factor A (VEGF-A), used as an anti-angiogenic agent in the treatment of various cancers[3]. - Chemotherapy, referring to standard-of-care cytotoxic agents, in Phase II metastatic solid tumor and metastatic colorectal cancer trials often mFOLFOX6 or FOLFIRI regimens[8]. The combination is under investigation for synergistic immunotherapy and anti-angiogenic effects in advanced/metastatic solid tumors, especially colorectal cancer, where immune checkpoint blockade alone is insufficient. Ongoing trials focus on metastatic colorectal cancer and other advanced solid tumors, aiming to overcome immune resistance by simultaneously inhibiting TGF-β, blocking PD-1, suppressing angiogenesis, and cytotoxic tumor cell killing[8].
