nitazoxanide + ribavirin + hydroxychloroquine is a triple combination antiviral regimen, also known as TriACT (Triple Combination Antiviral Coronavirus Therapy), being evaluated for the treatment of COVID-19. Developed by Rutgers, The State University of New Jersey, in collaboration with Synavir Corporation, this therapy combines three small molecule drugs with distinct mechanisms of action: nitazoxanide (a thiazolide antiviral), ribavirin (a guanosine analog), and hydroxychloroquine sulfate (an aminoquinoline). The combination was designed based on in vitro studies demonstrating multiplicative antiviral effects against SARS-CoV-2 by targeting different stages of the viral replication life cycle. The regimen is typically administered as a 5-day oral course and has been evaluated in Phase 2 clinical trials to determine its efficacy in reducing viral load and preventing hospitalization in outpatients with early-stage COVID-19.
