Overview

The combination of nivolumab (a PD-1 immune checkpoint inhibitor) and BMS-986253 (an anti-interleukin 8 [IL-8] monoclonal antibody) is an investigational immunotherapy regimen for advanced cancers. Nivolumab blocks the PD-1 receptor, enhancing T-cell activity against tumors. BMS-986253 binds to free IL-8, preventing its signaling through CXCR1 and CXCR2 receptors, which may reduce recruitment of immunosuppressive myeloid-derived suppressor cells to the tumor microenvironment and improve immune-mediated tumor cell lysis. This combination aims to overcome resistance to checkpoint inhibitors by targeting both adaptive and innate immune escape mechanisms.
