Nivolumab + daratumumab is an investigational combination therapy consisting of the PD-1 checkpoint inhibitor nivolumab and the CD38-directed monoclonal antibody daratumumab. Developed through a clinical collaboration between **Bristol Myers Squibb** and **Janssen Biotech**, the combination aims to leverage synergistic immunomodulatory effects. Daratumumab targets CD38-expressing malignant cells and depletes CD38-positive immunosuppressive cells (such as myeloid-derived suppressor cells and regulatory T cells), which may overcome resistance to PD-1/PD-L1 blockade and enhance the T-cell mediated antitumor activity of nivolumab. The combination has been primarily evaluated for the treatment of relapsed or refractory multiple myeloma (RRMM) and various solid tumors, including non-small cell lung cancer and head and neck squamous cell carcinoma.
