Overview

Nivolumab + dasatinib is an investigational combination of two distinct anti-cancer agents—nivolumab, a human IgG4 monoclonal antibody immune checkpoint inhibitor targeting programmed death-1 (PD-1) receptor, and dasatinib, an oral small-molecule tyrosine kinase inhibitor that inhibits multiple kinases including BCR-ABL and Src family kinases. The rationale for the combination was to potentially increase anti-leukemic activity in chronic myeloid leukemia (CML) by simultaneously enhancing antitumor immune response (nivolumab) and inhibiting oncogenic tyrosine kinase signaling (dasatinib). The combination has demonstrated safety in early-phase trials in CML, but did not show significant clinical activity in this setting, and development appears limited to early-phase investigation for hematologic malignancies[1][5][7][9].
