A quadruplet combination regimen consisting of **nivolumab** (anti-PD-1 monoclonal antibody immune checkpoint inhibitor), **elotuzumab** (anti-SLAMF7 monoclonal antibody activating NK cells and enhancing antibody-dependent cell-mediated cytotoxicity), **pomalidomide** (oral immunomodulatory agent enhancing immune responses and direct cancer cell toxicity), and **dexamethasone** (synthetic glucocorticoid corticosteroid with anti-inflammatory and anti-proliferative effects). This regimen was explored for treatment of **relapsed or refractory multiple myeloma** in clinical trials. The underlying rationale is that pomalidomide can sensitize myeloma cells to immune-mediated attack, dexamethasone supports control of inflammation, elotuzumab enhances NK cell and monoclonal antibody cytotoxicity, while nivolumab augments cytotoxic T cell activity by blocking PD-1 checkpoint signaling, collectively seeking synergistic anti-myeloma effects[1][3][5][7]. Trials indicated modest or no added benefit over standard combinations, and development was terminated.
