Overview

This is a combination regimen consisting of three agents: **nivolumab** (a monoclonal antibody targeting programmed death-1 [PD-1]), **ipilimumab** (a monoclonal antibody targeting cytotoxic T-lymphocyte-associated protein 4 [CTLA-4]), and **cyclophosphamide** (an alkylating chemotherapeutic agent). Nivolumab and ipilimumab are recombinant human monoclonal antibodies of the IgG4 (nivolumab) and IgG1 (ipilimumab) subclasses, respectively, and are immune checkpoint inhibitors that enhance antitumor immune responses via T-cell activation. Cyclophosphamide is a synthetic alkylating agent belonging to the oxazaphosphorine group, used to suppress regulatory T cells and potentiate immune responses or as a cytotoxic agent. This combination has been studied primarily in advanced/metastatic cancers (notably breast cancer) within early-phase clinical trials. The regimen is under clinical investigation and is not an approved standard of care for any disease.
