This is an experimental combination therapy consisting of **nivolumab** (a PD-1 checkpoint inhibitor monoclonal antibody), **ipilimumab** (a CTLA-4 checkpoint inhibitor monoclonal antibody), and **trametinib** (a small molecule MEK inhibitor). Nivolumab and ipilimumab work synergistically to activate the immune system against tumor cells by blocking inhibitory signals on T cells, while trametinib inhibits the MAPK/ERK pathway by blocking MEK1/2, thereby suppressing tumor proliferation commonly driven by BRAF mutations. This combination aims to maximize antitumor immune response while targeting oncogenic pathways in cancers, primarily developed and trialed in advanced melanoma, particularly with BRAF mutations.
