Overview

A combination regimen of **nivolumab** (a monoclonal antibody immune checkpoint inhibitor targeting PD-1), **pomalidomide** (an oral immunomodulatory drug), and **dexamethasone** (a synthetic glucocorticoid corticosteroid). This triplet is mainly studied for the treatment of **relapsed and refractory multiple myeloma (RRMM)**, particularly in patients who have failed prior lines of therapy including immunomodulators and proteasome inhibitors. Nivolumab enhances anti-tumor T-cell responses by blocking the inhibitory PD-1 pathway; pomalidomide induces direct tumoricidal effects and modulates innate and adaptive immune responses; dexamethasone exerts anti-inflammatory and antitumor activities. The combination is being assessed in clinical trials (e.g., CheckMate 602, NCT02726581) and is not currently approved for routine clinical use.
