Nivolumab + sitravatinib is an investigational combination therapy for cancer, pairing two distinct agents with complementary mechanisms. Nivolumab is a monoclonal antibody that inhibits the programmed cell death protein 1 (PD-1) receptor, thereby enhancing T-cell–mediated immune responses against tumors. Sitravatinib is an oral small molecule tyrosine kinase inhibitor targeting multiple receptors including TYRO3, AXL, and MERTK (collectively known as TAM receptors), as well as VEGFR2 and MET. By inhibiting these kinases, sitravatinib reduces immunosuppressive myeloid cells in the tumor microenvironment and may resensitize tumors to immune checkpoint blockade. The combination has been studied primarily in advanced/metastatic non-small cell lung cancer (NSCLC), clear cell renal cell carcinoma (ccRCC), and urothelial carcinoma after progression on prior therapies including checkpoint inhibitors or chemotherapy[1][3][4][5][6]. Clinical trials have shown manageable safety profiles but mixed efficacy results depending on tumor type and patient population.
