Overview

Nivolumab + tesetaxel is an investigational combination therapy consisting of two anti-cancer agents with distinct mechanisms of action. Nivolumab is a fully human monoclonal antibody that targets and inhibits the programmed death 1 (PD-1) receptor on T cells, thereby blocking the interaction with its ligands PD-L1 and PD-L2. This immune checkpoint inhibition enhances T-cell mediated anti-tumor responses by preventing tumor-induced immune suppression. Tesetaxel is an orally administered taxane-class small molecule that disrupts microtubule function, inhibiting cell division and inducing apoptosis in cancer cells. The combination has been studied in clinical trials for advanced solid tumors, including metastatic triple-negative breast cancer (TNBC), where it aims to leverage both immunotherapeutic and cytotoxic effects[3][4][7].
