Overview

This combination therapy involves the programmed death-1 (PD-1) immune checkpoint inhibitor nivolumab administered in conjunction with transarterial chemoembolization (TACE). Nivolumab is a human monoclonal antibody that blocks the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby enhancing T-cell mediated anti-tumor immune responses. TACE is a localized treatment for hepatocellular carcinoma that involves the delivery of chemotherapy (often via drug-eluting beads) directly to the tumor's blood supply followed by embolization to induce ischemia. The combination is being investigated to determine if the local inflammatory environment and antigen release caused by TACE can synergize with the systemic immune activation of nivolumab in patients with intermediate-stage hepatocellular carcinoma.
