Overview

Nivolumab is a monoclonal antibody that targets the programmed death-1 (PD-1) receptor on T cells and acts as an immune checkpoint inhibitor to enhance anti-tumor immune responses. Tumor-infiltrating lymphocytes (TILs) are autologous immune cells isolated from a patient's tumor and expanded ex vivo for adoptive cell therapy; they are reinfused to mediate direct anti-tumor effects. Interferon-alpha (IFN-alpha) is a cytokine with immunomodulatory properties that can recruit effector T cells to tumor sites and potentially enhance the efficacy of immunotherapies. The combination of these three modalities aims to synergistically boost anti-tumor immunity by blocking PD-1-mediated inhibition (nivolumab), providing cytotoxic effector cells (TILs), and enhancing immune cell recruitment/activation at the tumor site (interferon-alpha). This approach has been investigated in early-phase clinical trials for advanced solid tumors such as melanoma and non-small cell lung cancer[1][2][3][4][5].
