Overview

**NK:BOB1-TCR** is an engineered cell therapy where human primary natural killer (NK) cells are genetically modified to express a T cell receptor (TCR) specific for the B cell-specific transcription factor BOB1, presented in the context of HLA-B*07:02. This product may include co-expression of additional proteins, such as CD8 and CD3 chains, to support TCR functionality in NK cells. BOB1 is highly expressed in both healthy and malignant B cell lineages, including multiple myeloma. The addition of the BOB1-TCR enables NK cells to recognize and kill BOB1+, HLA-B*07:02+ tumor cells with HLA-dependent, antigen-specific cytotoxicity. Mechanistic studies have demonstrated potent anti-tumor effects in vitro and delay in tumor outgrowth and improved survival in preclinical mouse models of multiple myeloma. Some versions of the product may also be engineered to autonomously secrete IL-15, further enhancing both NK- and TCR-mediated cytotoxicity and persistence in vivo. This modality represents a novel approach combining the cytotoxic properties of NK cells and the antigen specificity of TCRs for adoptive cellular immunotherapy in B-cell malignancies, especially multiple myeloma[1][3][5][7].
