Overview

This therapeutic regimen, developed by the National Cancer Institute (NCI), involves the infusion of donor-derived, activated natural killer (NK) cells following an allogeneic peripheral blood stem cell transplant (HSCT). It is specifically investigated for pediatric and young adult patients with high-risk hematologic malignancies, such as leukemia and lymphoma, as well as refractory solid tumors. The approach aims to leverage the innate anti-tumor activity of NK cells to eliminate residual cancer cells (minimal residual disease) and prevent relapse after the transplant. The NK cells are collected from the same donor as the stem cells and are typically activated ex vivo before being infused into the recipient at specific intervals post-transplant. The goal is to enhance the graft-versus-tumor effect without increasing the risk of graft-versus-host disease (GvHD).
