Overview

NK cells + nogapendekin alfa is an investigational combination immunotherapy consisting of adoptively transferred universal donor (non-HLA matched) natural killer (NK) cells and the interleukin-15 (IL-15) superagonist ALT-803 (nogapendekin alfa). ALT-803 is a recombinant fusion protein complex designed to mimic the biological activity of IL-15 trans-presentation; it consists of an IL-15 mutant (N72D) with increased biological activity, bound to the sushi domain of the IL-15 receptor alpha (IL-15Rα) and fused to a human IgG1 Fc domain. This structure enhances the half-life and potency of the cytokine. In clinical trials led by Brenda Cooper, MD, ALT-803 is administered following NK cell infusion to support the survival, expansion, and anti-tumor cytotoxicity of the donor cells. The combination is being evaluated for the treatment of various refractory hematologic malignancies, such as acute myeloid leukemia and multiple myeloma, as well as advanced solid tumors including colon carcinoma and sarcomas.
