This is a combination regimen of three investigational and/or approved agents: - **NKT2152** is a potent, selective, orally available small molecule inhibitor of HIF2α (hypoxia-inducible factor 2-alpha), designed to prevent transcription of HIF2α-regulated genes involved in tumorigenesis, angiogenesis, and glycolysis. It is in development for clear cell renal cell carcinoma (ccRCC)[3][5][7]. - **Palbociclib** is an approved small molecule cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor that blocks cell cycle progression, thereby inhibiting tumor cell proliferation. It is used for hormone receptor-positive breast cancers and is under investigation for other cancers[2][4][6]. - **Sasanlimab** is an investigational monoclonal antibody targeting PD-1 (programmed cell death protein 1), functioning as an immune checkpoint inhibitor to enhance anti-tumor T-cell response. This triplet combination is currently being evaluated in a phase 2, open-label, global clinical trial for patients with advanced or metastatic clear cell renal cell carcinoma who have progressed after prior VEGF/VEGFR therapy and at least one immune checkpoint inhibitor[5][7]. The goal is to assess safety, efficacy, pharmacokinetics, and to determine the optimal dosing regimen for this combination in this cancer population.
