Overview

NKTR-255 + daratumumab is a combination investigational regimen consisting of NKTR-255, a PEGylated recombinant human interleukin-15 (IL-15) receptor agonist, and daratumumab, an anti-CD38 monoclonal antibody. NKTR-255 is designed to enhance the proliferation and activation of CD8+ T cells and natural killer (NK) cells through IL-15 receptor stimulation, thereby potentiating antitumor immune responses. Daratumumab binds CD38 on malignant plasma cells, mediating tumor cell death by antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity, and apoptosis. Preclinical and early clinical studies indicate that the combination augments NK cell-mediated ADCC and overcomes daratumumab-induced NK cell depletion, resulting in improved antitumor efficacy in multiple myeloma and potentially other hematologic malignancies.
