NKTR-262 + bempegaldesleukin is a **combination investigational immunotherapy for cancer** that couples a **small molecule toll-like receptor 7/8 agonist (NKTR-262)** with a **pegylated interleukin-2 (IL-2) pathway agonist (bempegaldesleukin, also called NKTR-214)**. NKTR-262 is designed for intratumoral injection to locally activate antigen-presenting cells and initiate a de novo cytotoxic T cell response. Bempegaldesleukin is administered systemically and acts as a CD122-biased agonist, promoting expansion and proliferation of CD8+ T cells and natural killer (NK) cells within the tumor microenvironment. The intended synergistic effect is to enhance both local and systemic antitumor immunity via coordinated activation of innate and adaptive immune responses. This combination was developed and trialed primarily by Nektar Therapeutics for the treatment of selected advanced/metastatic solid tumors, including melanoma, merkel cell carcinoma, triple negative breast cancer, renal cell carcinoma, colorectal cancer, head and neck squamous cell carcinoma, and sarcoma. The main program was terminated after phase 1, and development has not advanced further in clinical trials[1][3][4][5][6].
