NLM001 + zalifrelimab is an investigational combination therapy being developed by Nelum Corp in collaboration with Agenus for the treatment of first-line advanced pancreatic cancer. The regimen consists of NLM001 (also known as NLM-001), a small-molecule Hedgehog pathway inhibitor, and zalifrelimab (AGEN1884), a monoclonal antibody that acts as a CTLA-4 antagonist. NLM001 is designed to target cancer-associated fibroblasts within the tumor microenvironment, thereby promoting immune cell infiltration and improving the penetration of co-administered chemotherapy. Zalifrelimab serves as a checkpoint inhibitor to enhance the anti-tumor immune response. This combination is currently being evaluated alongside standard-of-care chemotherapy in the Phase 1/2 NUMANTIA clinical trial (NCT04827953).
