Overview

NMS-03305293 + temozolomide is an experimental combination therapy comprising NMS-03305293, an orally bioavailable, selective poly(ADP-ribose) polymerase 1 (PARP1) inhibitor, and temozolomide, an oral alkylating agent. NMS-03305293 selectively inhibits PARP1, disrupting DNA repair and inducing synthetic lethality in tumor cells with homologous recombination deficiency, notably BRCA1/2-mutated and other HRD tumors. Temozolomide induces DNA alkylation and damage, increasing cytotoxicity in rapidly dividing cells. The combination is being studied for synergistic antitumor effects, particularly in diffuse gliomas and glioblastoma, by exploiting the DNA repair defect targeted by NMS-03305293 and the cytotoxic DNA damage induced by temozolomide. The combination is under early clinical investigation primarily for brain tumors and other advanced solid tumors[8][9].
