Overview

This combination therapy, developed by ImmunityBio, Inc., consists of nogapendekin alfa-inbakicept (NAI; brand name ANKTIVA) and allogeneic invariant natural killer T (iNKT) cells (AgenT-797). It is currently being evaluated in Phase 2 clinical trials for the treatment of critically ill adults with severe community-acquired pneumonia (CAP) and lymphopenia, including cases complicated by sepsis or acute respiratory distress syndrome (ARDS). Nogapendekin alfa-inbakicept is a recombinant IL-15 superagonist complex (IL-15N72D:IL-15RαSu/IgG1 Fc) that stimulates the proliferation and activation of natural killer (NK) cells and CD8+ T cells by binding to the IL-15 receptor beta/gamma subunits. AgenT-797 is an off-the-shelf, donor-derived iNKT cell product that acts as an immune system orchestrator, bridging innate and adaptive immunity and providing immunomodulatory effects without the requirement for HLA matching. The combination is designed to restore immune function and address the severe lymphopenia often seen in patients with life-threatening respiratory infections.
