Overview

nogapendekin alfa inbakicept + Tri-Ad5 vaccines is a combination immunotherapy developed by ImmunityBio in collaboration with the National Cancer Institute (NCI). The regimen consists of nogapendekin alfa inbakicept (also known as N-803), an IL-15 superagonist fusion protein, and Tri-Ad5, a trivalent vaccine utilizing adenovirus type 5 (Ad5) vectors to deliver three tumor-associated antigens: Carcinoembryonic Antigen (CEA), Mucin-1 (MUC1), and Brachyury. N-803 is designed to enhance the proliferation and activation of natural killer (NK) cells and CD8+ T cells by mimicking the activity of IL-15, while the Tri-Ad5 vaccine primes the immune system to recognize and attack cells expressing the targeted antigens. This combination is currently being evaluated in Phase 2 clinical trials (such as QUILT-5.015) for the prevention and treatment of colorectal cancer in patients with Lynch syndrome.
