Nous-PEV + pembrolizumab is a combination therapy comprising a personalized viral vector-based neoantigen vaccine (NOUS-PEV) and pembrolizumab, a monoclonal antibody checkpoint inhibitor. NOUS-PEV is designed to target up to 60 patient-specific tumor neoantigens using a prime-boost strategy with adenoviral (GAd-PEV) and modified vaccinia Ankara (MVA-PEV) vectors, aiming to induce robust neoantigen-specific T-cell responses. Pembrolizumab blocks the interaction between PD-1 and its ligands, enabling stronger immune-mediated tumor cell killing. This combination has been studied primarily in patients with metastatic melanoma and non-small cell lung cancer (NSCLC) with high PD-L1 expression, to enhance anti-tumor immune response. Early studies suggest the combination is feasible, immunogenic, and has manageable toxicity.
