**NP-G2-044 + anti-PD-1 therapy** is a combination regimen involving NP-G2-044, a first-in-class small molecule fascin inhibitor, co-administered with a standard-of-care anti-PD-1 immune checkpoint inhibitor. NP-G2-044 inhibits fascin, an actin cross-linker key for filopodia formation and cancer cell metastasis, impeding tumor invasiveness and increasing dendritic cell-mediated antigen uptake. Anti-PD-1 therapies work by blocking programmed death-1 (PD-1) signaling on T cells, reactivating an immune response against tumors. This combination has demonstrated durable responses and tumor control in patients with advanced or metastatic solid tumors, particularly those resistant to prior anti-PD-1 therapies. NP-G2-044 is developed and manufactured by Novita Pharmaceuticals and has entered phase 2 clinical studies, showing promising disease control and objective response rates across various cancer types, including cervical, endometrial, pancreatic, gastroesophageal junction adenocarcinoma, cutaneous squamous cell carcinoma, non-small cell lung cancer, and cholangiocarcinoma[1][3][5].
