NP137 + paclitaxel + carboplatin is a combination therapy consisting of three agents: NP137, paclitaxel, and carboplatin, under investigation primarily for the treatment of advanced or metastatic endometrial and cervical cancers after prior chemotherapy. NP137 is a humanized monoclonal IgG1 antibody targeting the extracellular protein netrin-1, which is overexpressed in various cancers and involved in promoting tumor survival, epithelial-mesenchymal transition (EMT), metastasis, and resistance to therapy. By neutralizing netrin-1, NP137 blocks its interaction with dependence receptors (such as UNC5B and DCC), thereby restoring apoptosis in tumor cells and inhibiting EMT. Paclitaxel and carboplatin are established cytotoxic chemotherapies that function via microtubule stabilization and DNA crosslinking, respectively, leading to apoptosis of rapidly dividing cells. The combination aims to enhance antitumor efficacy, improve survival, and overcome chemotherapy resistance by integrating targeted therapy (NP137) with standard chemotherapy (paclitaxel and carboplatin)[1][2][3][5][6].
