NPE + UCHT1 + folate is an **engineered conjugate** consisting of the monoclonal antibody UCHT1 (anti-human CD3), chemically modified by the addition of folate groups (enabling targeting to folate receptor-expressing tumor cells) and further coated with NPE (2-nitrophenylethyl) groups, which **reversibly inactivate the antibody's activity until exposure to UV light**. The folation allows the conjugate to bind to folate receptor-positive cancer cells, and the NPE coating suppresses T cell activation by sterically blocking the antigen-binding site. Upon UV activation, the NPE groups are cleaved, restoring anti-CD3 activity and triggering T-cell mediated immune effects locally at the tumor site. This system is an experimental approach to targeted **immunotherapy for cancer**, enabling control over antibody activity to limit off-tumor toxicity. Studies have demonstrated that NPE-coated, folate-conjugated UCHT1 can significantly reduce tumor growth in vivo (in murine models), particularly when UV light is used to reactivate antibody activity at the tumor site[1].
