Overview

A combination of a **nonsteroidal anti-inflammatory drug (NSAID)** and a **triptan** is used mainly for the **acute treatment of migraine**. The most well-studied and widely approved example is the fixed-dose combination of **sumatriptan** (a triptan, serotonin 5-HT₁B/₁D agonist) and **naproxen sodium** (an NSAID that inhibits cyclooxygenase-1 and -2 enzymes, reducing prostaglandin synthesis and inflammation). This combination provides superior efficacy for achieving sustained pain freedom and relief compared to monotherapy with either agent, as each component targets distinct pathophysiological mechanisms in migraine: triptans act on the trigeminovascular system to induce vasoconstriction and block pain signal transmission, while NSAIDs reduce neurogenic inflammation in the trigeminal ganglion and prevent central sensitization. Typical indications are moderate to severe episodic migraine in adults.
